Overview

Sorting nexin-4 (SNX4) is a member of the sorting nexin family characterized by a PX domain that binds phosphoinositides and a BAR domain that remodels membrane curvature. SNX4 acts as a central adaptor in the recycling pathway from endosomes to the cell surface and is involved in autophagosome assembly. It forms complexes with other sorting nexins (SNX7, SNX30, SNX5, SNX32) to mediate trafficking and protein homeostasis[1][2]. SNX4 is expressed in all cell types and is especially active in neurons, regulating synaptic vesicle docking and neurotransmission[2]. Dysregulation of SNX4 affects endosomal sorting and may be implicated in cancer progression and neurodegenerative diseases such as Alzheimer’s—where SNX4 aids in the recycling of the amyloid precursor protein cleaving enzyme BACE-1, preventing its mislocalization[2]. Altered SNX4 expression can serve as a biomarker for disease prognosis and immune infiltration, especially in renal carcinoma[1].
