Sorting nexin-5 (SNX5) is an endosomal trafficking protein that plays a critical role in the sorting and retrograde transport of membrane proteins from endosomes to the trans-Golgi network through its action as part of the retromer complex. SNX5 is distinguished by the presence of a PX domain, which binds specific phosphoinositides, and a BAR domain, which senses and induces membrane curvature. It is essential for the formation of macropinosomes derived from membrane ruffles in macrophages, participating in CSF-1 receptor-driven actin polymerization. SNX5 is also implicated in various pathological conditions including cancer and rare genetic diseases due to its role in cellular trafficking and protein complex assembly. Despite its importance in cell biology, SNX5 is not currently an established therapeutic target and no approved drugs interact with it directly
