Overview

SPAG5 antisense RNA 1 (SPAG5-AS1) is a cytoplasmic long non-coding RNA (lncRNA) that regulates the neighboring gene SPAG5 at both transcriptional and post-translational levels. It acts by sponging microRNAs (notably miR-769-5p), thereby modulating the expression of the transcription factor YY1, which in turn upregulates SPAG5. SPAG5-AS1 also binds to the deubiquitinase USP14 to stabilize SPAG5 protein. Functionally, SPAG5-AS1 is implicated in the regulation of cell apoptosis and autophagy, specifically in high-glucose–treated human podocyte models, via the SPAG5/AKT/mTOR signaling pathway. While it indirectly affects pathways and proteins recurrently implicated in cancer biology, including the regulation of the established oncogene SPAG5, SPAG5-AS1 itself is not currently established as a direct therapeutic target or approved biomarker.
