SPANXA2 overlapping transcript 1 (SPANXA2-OT1) is a long non-coding RNA (lncRNA) that is significantly upregulated in specific types of kidney injury, particularly crystalline nephropathy caused by calcium oxalate crystals. Experimental evidence shows it regulates the occurrence and development of epithelial-mesenchymal transition (EMT) in renal tubular epithelial cells by acting as a molecular sponge for miR-204, which in turn modulates the expression of Smad5. Through this axis, SPANXA2-OT1 promotes EMT, inhibits cell proliferation, and promotes apoptosis in renal epithelial cells. Additionally, it may influence chemokine expression and macrophage chemotaxis, suggesting a broader immunoregulatory role. Its dysregulation has also been noted in non-functioning pituitary adenomas and cancers, indicating possible wider relevance in oncology, but it is not characterized as a therapeutic target such as a receptor, enzyme, or transporter[1][3][4].
