Transcription factor Sp1 is a ubiquitously expressed zinc finger transcription factor encoded by the SP1 gene. It binds GC-rich motifs of many promoters throughout the human genome and regulates the transcription of numerous genes involved in essential processes such as cell differentiation, proliferation, apoptosis, immune responses, and chromatin remodeling. As a founding member of the Sp/Krüppel-like factor (Sp/KLF) family, Sp1 plays a critical role in both normal development and pathological states, especially cancer, where its overexpression is often associated with poor prognosis and the regulation of “hallmark” cancer genes involved in proliferation, immortality, angiogenesis, and immune evasion. Sp1 activity is highly regulated by post-translational modifications, and it can function as both an activator and a repressor of transcription depending on context. Due to its centrality in gene regulation and disease—particularly tumorigenesis—Sp1 is considered a potential but challenging therapeutic target; direct inhibitors are under investigation but none are widely approved for clinical use.
Inhibitors or modulators of Sp1 interfere with binding to GC-rich motifs in DNA, ultimately altering transcription of genes involved in cell growth, apoptosis, and other processes. Post-translational modifications (e.g., phosphorylation, acetylation, glycosylation) can modulate Sp1's activator or repressor functions, affecting downstream gene expression.
