Specificity protein 1 (SP1) is a ubiquitous zinc-finger transcription factor that binds to GC-rich motifs (GC-boxes) in the promoter regions of a wide variety of genes (Wikipedia, 2024). It belongs to the Sp/KLF family, which includes other members like SP2, SP3, and SP4 that also recognize similar DNA sequences and often compete for the same binding sites (NIH, 2015). SP1 plays a critical role in regulating essential cellular processes such as cell growth, differentiation, apoptosis, and angiogenesis by controlling the expression of both housekeeping and tissue-specific genes (Frontiers, 2023). In many cancers, SP1 is overexpressed and acts as a master regulator of oncogenes and pro-survival factors, making it a significant therapeutic target (AACR, 2018). Drugs like Mithramycin A (Plicamycin) target SP1 by binding to GC-rich DNA sequences and preventing transcription factor occupancy, while other agents like tolfenamic acid and betulinic acid induce the degradation of SP proteins (NIH, 2016). Biomarkers such as SP1 protein levels and the expression of downstream targets like survivin are often used to monitor the efficacy of SP1-targeted therapies (NIH, 2016). However, because SP1 is involved in the basal transcription of many essential genes, therapeutic targeting is often limited by systemic toxicity and a narrow therapeutic window (Safe, 2023).
Inhibition of DNA binding to GC-rich promoter regions, induction of proteasomal degradation of Sp proteins, or modulation of transcriptional activity through post-translational modifications.
