Overview

SYNE2 (Spectrin repeat containing nuclear envelope protein 2) encodes nesprin-2, a large, modular protein located in the outer nuclear membrane that binds cytoplasmic F-actin. Nesprin-2 is a key component of the LINC complex, which physically links the nuclear envelope to the cytoskeleton, thereby maintaining nuclear architecture and regulating nuclear movement in numerous cell types. This connection facilitates mechanotransduction, nuclear positioning, and migration. SYNE2's interactions with inner nuclear membrane proteins (such as emerin, lamins, SUN proteins), actin-binding proteins, and motor adaptors underpin its structural and regulatory functions. Pathogenic mutations in SYNE2 are implicated in autosomal dominant Emery-Dreifuss muscular dystrophy and nuclear envelope-related disorders, and genetic variation in SYNE2 can affect cancer outcomes
