Overview

Sperm acrosome-associated protein 9 (SPACA9) is a structural microtubule inner protein found in human multiciliated cells and spermatozoa. It binds to the lumenal surface of both singlet and doublet microtubules, contributing to distinctive striations and spiral structures that stabilize the microtubule lattice, particularly in sperm flagella and respiratory cilia. SPACA9 forms part of the TAILS complex at the distal tip of sperm flagella and interacts in conjunction with SAXO proteins. Its presence and distribution impact the integrity and stress resistance of axonemal microtubules. Although a mouse knockout lacks obvious reproductive or structural phenotypes, the protein appears essential for specific human cilia and sperm microtubule architecture. Its potential functions include microtubule stabilization and acrosome complex formation, but it is not considered a therapeutic target, classified disease gene, or biomarker at present.
