Overview

Sperm-associated antigen 16 protein (SPAG16) is a structural protein essential for the formation and function of the sperm flagellum and motile cilia in eukaryotic cells[1][5]. It encodes at least two major isoforms: SPAG16L, which is integral to the axonemal central apparatus responsible for sperm motility, and SPAG16S, which localizes to nuclear speckles in male germ cells and participates in gene expression regulation during spermatogenesis[2]. Both isoforms contain seven WD repeat domains, which mediate multimeric protein interactions, facilitating their roles in cytoskeletal and gene expression complexes[4][6]. Mutations or deficiency in SPAG16 disrupt sperm motility, leading to male infertility, and are implicated in primary ciliary dyskinesia, a disorder of motile cilia function. SPAG16 is not currently recognized as a therapeutic drug target, and no approved drugs act via this protein.
