Overview

Sperm-associated antigen 6 (SPAG6) is a structural protein that localizes to the central apparatus of the axoneme in cilia and flagella, especially in sperm tails. It contains eight contiguous armadillo repeats that mediate protein-protein interactions critical for cilia and flagellar architecture and motility. In mammals, SPAG6 is essential for proper formation, alignment, and function of motile cilia and sperm flagella, thus playing a crucial role in fertility and in maintaining respiratory, neurological, and reproductive health. Mutations or dysregulation in SPAG6 can result in a spectrum of ciliary dysfunctions, such as primary ciliary dyskinesia, infertility, hydrocephalus, and potential hearing and neuronal defects. Antibodies against SPAG6 are implicated in immunologic infertility, making this antigen a candidate for diagnostic or contraceptive research.
