Overview

Sperm-associated antigen 7 (SPAG7) is a highly conserved, nuclear and cytoplasmic protein that contains an R3H domain predicted to bind polynucleotides such as single-stranded DNA or RNA. SPAG7 is primarily expressed in male germ cells, specifically during acrosome development of spermatids, and is also found in other tissues like muscle and nerve. It is believed to play a crucial role in sperm acrosome formation, tail structure, sperm motility, and male fertility. Deficiency of SPAG7 in animal models results in growth restriction, muscle atrophy, obesity, disrupted mitochondrial function in muscle, and significant reproductive deficits. Abnormal expression of SPAG7, and its regulation by specific microRNAs, is associated with male infertility, and alterations in its level are correlated with poor sperm count and motility. SPAG7 is not a classic therapeutic target such as a receptor or enzyme, but may serve as a biomarker for infertility and related disorders.
