Overview

Sperm-associated antigen 9 (SPAG9) is a scaffold protein encoded by the SPAG9 gene, classified as a cancer-testis antigen. It structurally organizes mitogen-activated protein kinases (MAPKs) and mediates c-Jun N-terminal kinase (JNK) signaling, functioning as a key modulator in signal transduction. SPAG9 facilitates the formation of functional JNK signaling complexes, binds to kinesin-1, and is implicated in microtubule-based transport, which may contribute to tumor growth, invasion, and development. It is highly expressed in several cancers (lung, cervical, bladder) and testicular germ cells, and is recognized as an autoantigen, provoking a strong humoral immune response in cancer patients. SPAG9 expression and related serum autoantibodies have shown promise as biomarkers for cancer diagnosis and patient monitoring[1][2][3][4][6].
