Overview

Sperm DNA integrity refers to the structural and chemical continuity of the genetic material within the male gamete, which is essential for successful fertilization and healthy embryo development. Unlike somatic cells, sperm DNA is uniquely packaged with protamines instead of histones to ensure extreme compaction and protection against environmental stressors during transit. Loss of integrity, often measured as sperm DNA fragmentation (SDF), arises from various factors including oxidative stress, abortive apoptosis, and defective chromatin remodeling during spermiogenesis. While not a single protein or receptor target, sperm DNA integrity is a critical clinical parameter and biomarker in reproductive medicine, particularly for diagnosing male-factor infertility that may not be apparent on a standard semen analysis. Therapeutic approaches do not target the DNA directly with drugs but rather focus on mitigating the underlying causes of damage, such as using antioxidants to neutralize reactive oxygen species or surgically treating varicoceles to reduce testicular heat and oxidative stress.
