Sperm equatorial segment protein 1 (SPESP1) is a protein highly expressed in the testis, primarily localized to the acrosome of postmeiotic spermatids and mature sperm, and concentrated in the equatorial segment of mature sperm. It is a membrane-associated protein essential for the fusion of sperm and egg plasma membranes during fertilization. SPESP1-deficient male mice exhibit significantly reduced fertility due to impaired sperm migration, decreased sperm-egg fusion, and abnormal distribution of acrosomal proteins. SPESP1 serves as an alloantigen in humans and plays a crucial role in the fertilization process. It is not currently considered a therapeutic target, and there are no known drugs that interact with it[1][2][4][5][6].
