Overview

Sperm microtubule inner protein 8 (SPMIP8) is a cytoskeletal protein specifically expressed in testis, prostate, and placenta, and conserved across chordate species[1][2][3][4][8]. It is localized to the flagellum axoneme of sperm, where it serves as a microtubule inner protein (MIP), possibly reinforcing and stabilizing the doublet microtubules in the axoneme and supporting motile cilium function[2][9]. Scientifically, SPMIP8 is a protein-coding gene with tissue-restricted expression suggestive of specialized roles in the structural integrity and motility of sperm flagella. Its expression is selective to reproductive tissues and it may contribute to reproductive biology, but current evidence does not support a specific enzymatic, receptor, or transport function[1][2][4][8]. There are no disease-modifying or therapeutic roles established for SPMIP8, nor are there direct pharmacological modulators, clinical biomarkers, or safety challenges documented in the literature.
