Sperm protein 17 (Sp17) is a highly conserved mammalian protein originally identified on the sperm flagellar fibrous sheath, where it is implicated in sperm maturation, fertilization, and sperm-oocyte binding[1][3]. Sp17 is now recognized to be widely distributed in both germinal (reproductive) and various somatic tissues, and is overexpressed in multiple human cancers, leading to its classification as a cancer/testis antigen (CTA)[2][5]. It contains domains homologous to protein kinase A anchoring protein and calmodulin-binding sites, and participates in protein-protein interactions important for cell signaling and motility in both sperm and cancer cells[1][2]. Its aberrant expression in tumors and functional involvement in cell migration/invasion makes Sp17 a candidate diagnostic and immunotherapeutic target, though safety and specificity remain important considerations[2][4][5].
Inhibiting cell surface Sp17 aims to block immune escape or tumor cell migration. Vaccine strategies are in preclinical/early clinical testing to stimulate antitumor immunity via recognition of Sp17-expressing tumor cells
