Sperm protein associated with the nucleus on the X chromosome A1 (SPANXA1) is a member of the SPANX gene family, which encodes cancer/testis antigens (CTAs) found mainly in testis and a range of tumor types but not in other normal tissues[1][2][3][4]. SPANXA1 localizes to the nucleus in spermatozoa, where it associates with nuclear vacuoles and the redundant nuclear envelope, contributing to sperm nuclear architecture and potentially regulating chromatin organization and nuclear processes[4][6][7]. In cancer, high SPANXA1 expression can modulate cell migration and invasion, suppressing epithelial-mesenchymal transition (EMT) and acting as a negative regulator of metastasis in experimental models[2][3]. Its restricted expression pattern in normal tissue and upregulation in tumors make it a potential immunotherapy target and a marker for cancer diagnosis and prognosis, as well as a marker for specialized nuclear structures in sperm[3][4][6].
