Overview

Sperm protein associated with the nucleus on the X chromosome D (SPANXD) is a member of the SPANX gene family, encoding small, highly homologous nuclear proteins specifically restricted to post-meiotic spermatids and mature spermatozoa in normal tissues, but aberrantly expressed in various cancers as cancer/testis antigens[1][2][3][5][6]. The SPANX-D isoform is part of the SPANX-A/D subfamily, which plays a multifunctional role in the nuclear envelope organization of sperm, regulates chromatin condensation, and is involved in mitochondrial energy metabolism and sperm motility[3][6]. SPANX-A/D proteins are also identified in several malignancies, promoting pro-tumoural processes like cell proliferation, motility, survival, invasion, and metastasis[3][5]. The restricted normal tissue distribution and immunogenic properties make SPANX-D and its family members attractive targets for cancer immunotherapy and biomarkers for tumor cell detection, with roles in male fertility assessment. No drugs are currently known to specifically target SPANXD, and specific mechanisms of drug action have not been described. Safety considerations include the potential for off-target effects on testicular function during immunotherapeutic intervention targeting SPANXD[3][5][6].
