Overview

SPANXN5 (Sperm protein associated with the nucleus on the X chromosome N5) is part of the SPANX (Sperm protein associated with nucleus, X chromosome) gene family, which encodes small nuclear proteins classified as cancer/testis antigens (CTAs). The family, including SPANXN5, is located on the X chromosome and exhibits highly restricted expression, being normally present only in testis but aberrantly activated in various cancers such as melanoma and breast cancers[2][4][7][8]. Functionally, SPANX proteins are involved in nuclear processes including chromatin organization, RNA binding and processing, and protein translation. In tumors, expression of SPANX proteins is associated with enhanced cell proliferation, migration, invasion, and metastasis, acting through interactions with nuclear proteins such as Lamin A/C[1][8]. Their tumor-restricted expression and immunogenic properties attract interest for cancer immunotherapy, but no direct small molecule or antibody drug interactions for SPANXN5 are documented; its clinical exploitation is mainly as a biomarker or potential target for immunotherapies[2][8].
