Overview

Spermadhesins are a family of secreted proteins characterized by the presence of a CUB domain and found predominantly in the seminal plasma of ungulates, such as pigs, cattle, and horses. They are multifunctional, peripherally bind to the sperm surface, and play a role in sperm capacitation and fertilization by mediating binding to carbohydrates and other ligands on the egg or reproductive tract mucosa. They have not been detected as functional genes in humans or rodents (most human and dog spermadhesin genes are inactive remnants), and are not linked to any validated therapeutic or disease targets. Spermadhesins are easily distinguished from other CUB domain-containing proteins, such as CUB domain-containing protein 1 (CDCP1), which are relevant in cancer biology and therapy. Spermadhesin family members are structurally and functionally distinct from other CUB domain-containing therapeutic targets. The term "SPADH" is not a canonical name for a therapeutic target in humans. There is no evidence they play any role as a drug target or biomarker in human medicine. Confusion may arise due to the overlap in naming with CUB domain proteins that *are* therapeutic targets (such as CDCP1). In this context, *SPADH* itself is not a drug target.
