Overview

Spermatogenesis and oogenesis specific basic helix-loop-helix-containing protein 1 (SOHLH1) is a germ cell-specific transcription factor belonging to the basic helix-loop-helix (bHLH) protein family[1][3]. It is preferentially expressed in oocytes and spermatogonia, and is essential for both female and male gametogenesis[1][3]. In females, SOHLH1 directs early folliculogenesis and is required for oocyte differentiation; its disruption leads to profound follicular defects and infertility due to loss of oocytes[1][3]. SOHLH1 acts by directly regulating the promoters of several other oocyte-specific transcription factors (including LHX8, ZP1, and ZP3), placing it upstream of critical regulators such as FIGLA and NOBOX in the genetic network controlling oogenesis[1][2]. In males, SOHLH1 is also required for spermatogonial differentiation, though its mechanisms are less well characterized. Mutations or deletions in SOHLH1 are linked to infertility syndromes caused by germ cell development failure, especially nonsyndromic ovarian insufficiency in women[1][3]. As a nuclear transcription factor, SOHLH1 does not currently have any known drug interactions or direct targeting by pharmacologic agents. Its clinical relevance primarily resides in its function as a genetic biomarker for certain infertility syndromes.
