Overview

SPATA12 is a protein coded by the SPATA12 gene, located on chromosome 3p14, with highly specific expression in the testis, notably in spermatocytes, spermatids, and spermatozoa. It is absent in supporting cells (Sertoli, Leydig) and fetal testis, with expression correlating to maturity and density of germ cells. SPATA12 functions as a negative regulator of cell proliferation during spermatogenesis, acting as a tumor suppressor by maintaining differentiated state and suppressing cell proliferation. It interacts with chromodomain helicase DNA binding protein 2 (CHD2) in the nucleus, participates in the DNA damage response, and is transcriptionally activated by AP-1 in response to oxidative stress or agents like resveratrol. SPATA12 also attenuates reactive oxygen species formation, malondialdehyde induction, and apoptosis under oxidative stress, underscoring its antioxidant role. Loss or reduction of SPATA12 expression is implicated in testicular germ cell tumors and male infertility. No established drugs target SPATA12 directly, although resveratrol can upregulate its expression.
