Overview

SPATA6 encodes a structural protein that plays a critical role in the development of the sperm connecting piece, linking the sperm head to the flagellum during late spermiogenesis[2]. This protein is highly conserved across vertebrates and is expressed exclusively in haploid germ cells (spermatids and spermatozoa)[1][2][3]. It is involved in microfilament transport via interactions with myosin subunits and is essential for male fertility; deficiency leads to acephalic spermatozoa (sperm without heads) and subsequent infertility[2][3]. In testicular germ cell tumor models, downregulation of SPATA6 is associated with reduced cell viability and enhanced apoptosis, suggesting a possible role in tumor cell survival and a potential target for cancer therapy[1]. No approved drugs directly target SPATA6, but its tissue- and development-specific expression makes it an important candidate for biomarker development in reproductive medicine[1][2][3].
