Overview

Spermatogenesis-associated serine-rich protein 1 (SPATS1) is an evolutionarily conserved, testis-specific protein highly expressed in primary spermatocytes during male meiotic prophase and scarcely expressed in other tissues[1][3][7]. It is a phosphoprotein thought to function in the regulation of germ cell differentiation and the early stages of spermatogenesis in mammals[3][7]. Research suggests it acts as a modulator of the canonical Wnt signaling pathway by interacting with Dishevelled 2 (DVL2) and promoting degradation of TCF4, potentially dampening Wnt pathway activity[1][5][7]. Mutations or downregulation of SPATS1 have been linked to impaired spermatogenesis and male infertility in humans, as well as possible risk for testicular cancer[1][5][7]. In mouse models, however, SPATS1 knockout did not impair fertility or spermatogenesis, indicating that it is not absolutely essential for these processes in all species[1][7]. There are currently no clinical drugs known to target SPATS1, and its primary significance lies in reproductive biology and as a possible biomarker for male reproductive health and certain cancers[1][3][5].
