Overview

The spermatozoal plasma membrane is a complex and highly organized lipid bilayer that serves as the outer boundary of the sperm cell, playing a fundamental role in the process of fertilization. It is responsible for regulating the transport of ions and metabolites, facilitating sperm-egg recognition, and undergoing the acrosome reaction necessary for penetrating the oocyte's protective layers [2]. Pharmacologically, the sperm membrane is the primary target for various spermicidal agents, most notably surfactants like nonoxynol-9 [1]. These drugs interact with the membrane to lower surface tension, causing physical disruption and lysis of the lipid bilayer, which leads to the immediate immobilization and death of the sperm cell [1, 4]. While effective as a contraceptive barrier, the non-specific action of these surfactants can also compromise the integrity of the vaginal and rectal mucosa, potentially increasing the risk of acquiring sexually transmitted infections such as HIV [3].

Mechanism of action

Surfactant-mediated disruption of the lipid bilayer, which increases membrane permeability, causes leakage of intracellular contents, and results in the rapid loss of sperm motility and viability [1, 4].
