Overview

Sphingomyelin phosphodiesterase 5 (SMPD5) is a mitochondria-associated neutral sphingomyelinase that catalyzes the hydrolysis of sphingomyelin to produce ceramide and phosphocholine, key intermediates in sphingolipid metabolism. This enzyme operates at neutral pH and is implicated in the regulation of mitochondrial lipid signaling, cell survival, and apoptosis. While alterations in sphingomyelinase activity are broadly implicated in neurodegenerative diseases and lysosomal storage disorders, direct roles for SMPD5 are under active investigation. No currently approved drugs specifically target SMPD5; however, its pathway is of increasing interest in understanding and potentially modulating lipid-related pathology[8][6][9].
