Overview

The sphingosine-1-phosphate (S1P) pathway is a critical lipid signaling cascade involved in numerous physiological and pathological processes, including immune cell trafficking, vascular development, and inflammation. S1P is a bioactive sphingolipid metabolite produced by the phosphorylation of sphingosine via two isoenzymes: sphingosine kinase 1 (Sphk1) and sphingosine kinase 2 (Sphk2). It exerts its effects primarily through five G protein-coupled receptors (S1PR1–5).
