Overview

Sphingosine 1-phosphate receptors (S1PRs) are a family of G protein-coupled receptors (GPCRs) that mediate the effects of the bioactive lipid sphingosine-1-phosphate (S1P). There are five known subtypes: S1PR1, S1PR2, S1PR3, S1PR4, and S1PR5. These receptors play crucial roles in regulating cell proliferation, migration, angiogenesis, immune cell trafficking, cytoskeleton organization, and mitogenesis. Modulators targeting these receptors are used for autoimmune diseases such as relapsing-remitting multiple sclerosis.

Modulators act as functional antagonists after initial agonist-induced internalization on lymphocytes, reducing circulating immune cells.
