It plays a crucial role in vascular maturation, barrier function, and immune cell trafficking. S1P1 is highly expressed on endothelial cells and lymphocytes. Its modulation is a validated therapeutic strategy for autoimmune diseases like multiple sclerosis.

Mechanism of action

Sphingosine 1-phosphate receptor modulators bind to S1PR1, leading to receptor internalization on lymphocytes and preventing lymphocyte egress from lymph nodes, thereby reducing circulating immune cells.
