Overview

Sphingosine-1-phosphate receptor 4 (S1PR4) is a G protein-coupled receptor that binds the lipid signaling molecule sphingosine-1-phosphate (S1P). It is primarily expressed on hematopoietic and lymphoid tissues, particularly immune cells like neutrophils and dendritic cells. S1PR4 plays a role in both innate and adaptive immunity, influencing cell proliferation, angiogenesis, migration, cytoskeleton organization, and immune cell trafficking. It couples primarily with Gi and G12/13 proteins, activating Rho kinase and influencing cytoskeletal rearrangement. Its selective expression on immune cells makes it a potential target for modulating immune responses, especially in autoimmune or inflammatory diseases.
