Overview

Sphingosine-1-phosphate receptor 5 (S1P5) is a G protein-coupled receptor that binds the lipid signaling molecule sphingosine-1-phosphate (S1P). It plays a crucial role in immune cell trafficking, central nervous system function, and lymphocyte regulation. S1P5 primarily signals through Gαi and G12/13 subunits, leading to downstream effects on cell migration, cytoskeleton organization, and immune cell trafficking. It is expressed in tissues such as the spleen, natural killer cells, patrolling monocytes, certain T cell subsets and oligodendrocytes within the CNS, where it is involved in remyelination processes relevant for diseases like multiple sclerosis. S1P5 influences differentiation of effector/memory T cells, regulating tissue residency during immune responses.
