Overview

Sphingosine 1-phosphate receptor subtype 1 (S1PR1) is a G protein-coupled receptor (GPCR) that plays a central role in vascular biology and immune system regulation. It regulates endothelial cell cytoskeletal structure, migration, capillary-like network formation, and vascular maturation. It also controls lymphocyte maturation, migration, trafficking—especially egress of T and B cells from thymus and secondary lymphoid tissues. Furthermore, it maintains vascular integrity and acts as mechanotransduction signaling in response to blood flow. Several drugs targeting this axis are approved/under development for autoimmune diseases such as multiple sclerosis (e.g., fingolimod), inflammatory bowel disease; preclinical evidence supports potential benefit in lupus erythematosus, rheumatoid arthritis, systemic sclerosis.
