Overview

Sphingosine 1-phosphate receptor subtype 3 (S1PR3) is a G protein-coupled receptor (GPCR) that binds the bioactive lipid sphingosine-1-phosphate (S1P). It plays a crucial role in regulating angiogenesis, immune responses, and hematopoietic stem cell fate. S1PR3 is implicated in various diseases including sepsis, acute myeloid leukemia, and intervertebral disc degeneration. Upon binding S1P, S1PR3 activates multiple downstream signaling pathways via coupling to different G proteins, influencing diverse cellular responses including inflammation, cell survival, migration, and differentiation. Its expression is inducible in hematopoietic stem cells upon exposure to pro-inflammatory cytokines like TNFα.
