Sphingosine 1-Phosphate Receptor Subtype 4 (S1PR4) is a G protein-coupled receptor that binds sphingosine 1-phosphate (S1P). It is primarily expressed in immune cells and brain endothelial cells. S1PR4 plays a role in immune cell trafficking, activation, and vascular permeability. It has been implicated in diseases such as asthma, cancer, and neurological disorders involving blood-brain barrier dysfunction, making it a potential therapeutic target.
Agonists bind to S1PR4, activating G proteins (Gαi/Gα12/13) and downstream signaling pathways such as MAPK/JNK and RhoA/ROCK, influencing immune cell function and vascular permeability.
