Sphingosine phosphate receptor 1 (S1PR1) is a G protein-coupled receptor that binds sphingosine-1-phosphate (S1P). It plays a crucial role in immune cell trafficking, vascular regulation, and cellular processes like proliferation and migration. S1PR1 is a validated therapeutic target for autoimmune diseases, particularly multiple sclerosis, due to its role in controlling lymphocyte egress from lymphoid tissues.
Binding to lymphocyte-expressed S1PR1 leads to internalization and functional antagonism, trapping lymphocytes within lymph nodes and preventing their egress into circulation.
