Overview

Spi-C transcription factor (SPIC) is an ETS-family transcriptional regulator, expressed primarily in B lymphocytes and red pulp macrophages. It plays a critical role in the development of red pulp macrophages necessary for iron homeostasis, and in the recycling of red blood cells. In B cells, Spi-C acts as a transcriptional repressor and may antagonize the activities of PU.1 and Spi-B, both of which are key drivers of B cell differentiation and antibody-mediated immune responses. It is also involved in the differentiation and function of osteoclasts, promoting bone resorption and responding to signals such as RANKL. Mutation or deletion of Spic affects immune cell development, and deficiencies can be associated with combined immunodeficiency syndromes. It is not currently considered a direct therapeutic target and no interacting drugs or defined safety concerns are reported.
