Overview

The spike glycoprotein (S protein) is a major structural component of the MERS-CoV virion, forming large protruding spikes on the virus surface. It is essential for viral entry into host cells and determines host specificity and infectivity. The S protein is a homotrimeric class I fusion protein, with each monomer composed of two subunits: S1 (receptor binding) and S2 (membrane fusion). The S1 subunit contains the receptor-binding domain (RBD) which specifically binds to dipeptidyl peptidase 4 (DPP4). The S protein undergoes proteolytic cleavage at S1/S2 and S2' sites. Mutations in the RBD can affect DPP4 affinity and antibody neutralization sensitivity. It is a major therapeutic target and diagnostic antigen.
