The spike glycoprotein (S protein) of SARS-CoV-2 is a large, trimeric, class I fusion protein found on the surface of the virus. It is essential for viral entry into host cells and is the primary target for neutralizing antibodies generated by infection or vaccination. The ancestral strain refers to the original Wuhan-Hu-1 sequence identified at the start of the COVID-19 pandemic. It mediates attachment to host cell receptors via its RBD. After binding ACE2, undergoes conformational changes that allow membrane fusion through its S2 subunit. Most first-generation COVID vaccines are based on either full-length or modified versions of this ancestral spike sequence.
