The spike glycoprotein (S protein) is a trimeric surface protein of the SARS-CoV-2 Delta variant that mediates viral entry into host cells. It binds to the human ACE2 receptor and facilitates membrane fusion. Key mutations in the Delta variant spike protein, such as L452R, T478K, and P681R, enhance ACE2 affinity, reduce antibody binding, and increase cleavage efficiency, leading to increased transmissibility and immune evasion. The spike protein is the primary target for neutralizing antibodies and vaccines.
Neutralizing antibodies bind to the spike protein, preventing its interaction with ACE2 and blocking viral entry. Vaccines elicit an immune response that generates antibodies targeting the spike protein.
