Overview

The spike glycoprotein (S) of SARS-CoV-2 contains the S1 subunit, which includes the receptor binding domain (RBD). The RBD is critical for recognizing and binding to human angiotensin-converting enzyme 2 (ACE2) receptors on host cells, initiating viral attachment and subsequent membrane fusion required for entry. The RBD is a major target for therapeutic antibodies and small-molecule inhibitors designed to block ACE2 interaction. It is also highly immunogenic, eliciting strong neutralizing antibody responses during natural infection or vaccination, making it a key target for vaccine development. Mutations in this region can significantly affect binding affinity, transmissibility, immune escape, and pathogenicity.
