Spinal cord neurons and peripheral nerve fibers constitute the fundamental structural and functional units of the somatosensory and motor systems. Spinal cord neurons, located within the central nervous system, integrate sensory input and coordinate motor output, while peripheral nerve fibers serve as the communication lines between the spinal cord and the rest of the body (StatPearls, 2023). These anatomical structures are not individual molecular targets but rather complex environments containing a multitude of specific therapeutic targets, including voltage-gated ion channels and various G protein-coupled receptors (PubMed, 2022). Dysfunction in these neurons and fibers is central to conditions such as neuropathic pain, spinal cord injury, and peripheral neuropathy (NIH, 2023). Drugs targeting these areas, such as analgesics and local anesthetics, typically work by modulating neuronal excitability or inhibiting signal propagation (PubChem, 2024). Consequently, while they are critical sites for pharmacological intervention, they represent a systemic level of organization rather than a discrete molecular entity.
Drugs acting on these structures typically modulate neuronal excitability through the inhibition of voltage-gated ion channels (e.g., Sodium or Calcium channels) or the activation/inhibition of neurotransmitter receptors (e.g., GABA or Opioid receptors) (PubMed, 2022; PubChem, 2024).
