Spindle apparatus coiled-coil protein 1 (SPDL1, Protein Spindly) is an adapter protein required for accurate chromosome segregation during cell division. It promotes dynein and dynactin localization to kinetochores and regulates spindle orientation and mitotic checkpoint signaling, thereby ensuring proper microtubule attachment and chromosome alignment. Mutations or altered expression of SPDL1 have been implicated in increased risk for idiopathic pulmonary fibrosis and some cancers, highlighting its biological and pathogenic significance. SPDL1 acts mechanistically by recruiting dynein/dynactin motors to kinetochores, facilitating the transition from lateral microtubule interaction to end-on attachment, and influencing cell migration by localizing to the leading edge of motile cells
