Spindlin interactor and repressor of chromatin-binding protein (SPINDOC, also known as C11orf84, SPIN-DOC, and SPIN1-docking protein) is a chromatin-associated protein that regulates the functions of the epigenetic reader Spindlin1. SPINDOC forms distinct protein complexes either with Spindlin1 or with PARP1, regulating pathways related to transcription and the DNA damage response[1][2][3]. By binding to Spindlin1, SPINDOC blocks Spindlin1’s ability to "read" the H3K4me3 histone mark, disassociating it from chromatin and acting as a transcriptional repressor[3]. In complex with Spindlin1, SPINDOC (C11orf84) plays a key role in recognizing non-canonical bivalent histone marks (H3K4me3K9me3) on rDNA loci, thereby regulating ribosomal RNA transcription and cell proliferation[2][4]. SPINDOC also directly interacts with PARP1 and promotes DNA repair-associated PARylation, indicating a role in genome maintenance under cellular stress[1]. While there is emerging evidence for SPINDOC’s roles in cancer and cell proliferation, SPINDOC is not currently considered a direct therapeutic target, and no known drugs interact directly with it.
