Spleen tyrosine kinase (SYK) is a cytoplasmic, non-receptor protein tyrosine kinase crucial for transducing signals from immune cell receptors, regulating cellular responses to antigens and immune complexes. It is primarily expressed in hematopoietic cells. SYK activation involves SH2 domain binding to phosphorylated ITAMs and subsequent autophosphorylation, leading to activation of downstream signaling pathways such as PI3K/Akt, Ras/ERK, PLCγ/NFAT, Vav/Rac, and IKK/NFκB. It plays critical roles in autoimmune diseases and cancer, making it a relevant therapeutic target. However, safety concerns exist regarding immunosuppression and off-target effects. SYK can also act as a tumor suppressor or promoter depending on the context.
