Overview

Spleen tyrosine kinase (SYK) recruitment to the Fc epsilon RI (FcεRI) gamma subunit is a critical step in mast cell and basophil activation following allergen exposure. Allergen-induced crosslinking of IgE bound to FcεRI leads to LYN-mediated phosphorylation of ITAMs on the γ subunits. These phosphorylated ITAMs then serve as docking sites for SYK, initiating downstream signaling cascades that result in degranulation and cytokine release. This pathway is central to allergic reactions.
