Splicing factor 3a subunit 1 (SF3A1) is a critical protein component of the SF3a heterotrimer within the spliceosome, specifically associated with the U2 snRNP. This protein acts as a scaffold and nuclear import determinant for the SF3a complex, facilitating assembly and stability. SF3A1 contains tandem SURP domains which mediate its interactions with SF3A2 and SF3A3, enabling the bridging of the 5′ and 3′ splice sites of pre-mRNA via interactions with U1 snRNA and U2 snRNA. Its C-terminal region supports nuclear localization and features a ubiquitin-like domain important for protein interactions. SF3A1 is essential for pre-mRNA splicing and alternative splicing, playing roles not only in spliceosome assembly but also in chromatin association and cell cycle regulation. Aberrant expression or mutation of SF3A1 is implicated in tumorigenesis, certain immune and infectious diseases, and may serve as a diagnostic or therapeutic target in oncology and immunology, with azacitidine being a cited interacting drug in myelodysplasia and related malignancies
