Splicing factor 3B subunit 1 (SF3B1) is a core component of the spliceosome, specifically part of the splicing factor 3b protein complex. It plays a critical role in pre-mRNA splicing, which is essential for generating mature messenger RNA from precursor transcripts. Mutations in SF3B1 are recurrently observed in several cancers, including myelodysplastic syndromes, chronic lymphocytic leukemia, and breast cancer. It is also involved in ferroptosis resistance. SF3B1 is therefore a therapeutic target in cancer.
SF3B1 inhibitors are being developed to disrupt splicing in cancer cells, leading to cell death. Specific mechanisms depend on the inhibitor.
