Overview

Splicing factor 3b subunit 5 (SF3B5) is a highly conserved protein component of the SF3B complex, which is part of the U2 small nuclear ribonucleoprotein (snRNP) involved in pre-mRNA splicing within the spliceosome[3][6]. SF3B5 plays a key role in stabilizing the interaction between U2 snRNA and the branch site of pre-mRNA, a critical step for accurate splice site selection and mRNA processing[3][6]. In addition to its core function in RNA splicing, SF3B5 has been shown in metazoans (e.g., Drosophila) to act as a subunit of the Spt-Ada-Gcn5 acetyltransferase (SAGA) transcriptional coactivator complex, where it mediates gene expression activation independently of its splicing role[1][2][4]. SF3B5 is essential for organismal development and cell viability; disruption can cause embryonic lethality and transcriptional defects, and is associated with specific human diseases such as certain congenital syndromes and cancer[6]. There are currently no known direct drugs or clinical biomarkers associated specifically with SF3B5.
