Sprouty-related, EVH1 domain-containing protein 1 (SPRED1) is a protein encoded by the SPRED1 gene on human chromosome 15q13.2. It belongs to the Sprouty family, acts as a tyrosine kinase substrate, and negatively regulates the Ras/MAPK signaling pathway by inhibiting Raf activation, thereby controlling MAP kinase cascade activation in response to growth factors. It can form homodimers or heterodimers with SPRED2, and is involved in processes like hematopoiesis, fibroblast growth factor signaling, actin stress fiber formation, and epithelial-to-mesenchymal transition. Mutations cause Legius syndrome (neurofibromatosis type 1-like syndrome) and are associated with childhood leukemia.
