SPRED3 is characterized by an N-terminal EVH1 domain and a C-terminal Sprouty-related (SPR) domain, but lacks the central c-Kit binding domain found in related family members, resulting in comparatively weaker inhibition of MAPK/ERK signaling[2][3]. Highly expressed in the brain and certain tumors, SPRED3 modulates growth factor signaling, influences cell proliferation, and inhibits processes like FGF-induced differentiation and TGF-beta-induced epithelial-to-mesenchymal transition[1][2][3]. Recent studies associate its overexpression with aggressive features and poor prognosis in various cancers, notably thyroid carcinoma, suggesting potential utility as a diagnostic or prognostic biomarker and possible therapeutic target[2][5].
